Theoretically, papain might increase the effects and side effects of warfarin.
In one case report, a patient previously stable on warfarin was found to have an international normalization ratio (INR) of 7.4, which was attributed to ingestion of a supplement containing papain from papaya extract.

Papain Overview
Papain is a proteolytic enzyme that is extracted from the papaya fruit. It is also known as papaya proteinase. Proteolytic enzymes are enzymes that break down proteins into smaller peptides and amino acids. Papain specifically breaks down proteins by hydrolyzing the peptide bonds that link amino acids together. Papain is used in a variety of applications, including food production, medicine, and biotechnology.
Medically, papain can be used to remove dead tissue from wounds and to treat certain skin conditions. It has also been used to treat sports injuries, such as sprains and strains. Additionally, it is a common ingredient in digestive enzyme dietary supplements to aid in the digestion of proteins.

Warfarin is used to prevent blood clots from forming or growing larger in your blood and blood vessels. It is prescribed for people with certain types of irregular heartbeat, people with prosthetic (replacement or mechanical) heart valves, and people who have suffered a heart attack. Warfarin is also used to treat or prevent venous thrombosis (swelling and blood clot in a vein) and pulmonary embolism (a blood clot in the lung). Warfarin is in a class of medications called anticoagulants ('blood thinners'). It works by decreasing the clotting ability of the blood.
