An increased risk of QT interval prolongation


Interaction Summary

Both azithromycin and nilotinib are known to prolong the QT interval[1]. Due to the potential for additive effects on the QT interval and increased risk of torsade de pointes, concomitant use of azithromycin and nilotinib should be avoided. However, if coadministration is required, the patient should be closely monitored for prolongation of the QT interval [2].

How To Manage Interaction

Coadministration of azithromycin and nilotinib, both drugs that prolong the QT interval, should be avoided due to the potential for additive effects on the QT interval and increased risk of torsade de pointes. However, if concurrent therapy is required, monitor patient closely for prolongation of the QT interval[2].

Azithromycin Overview

  • Azithromycin is used to treat certain bacterial infections, such as bronchitis; pneumonia; sexually transmitted diseases (STD); and infections of the ears, lungs, sinuses, skin, throat, and reproductive organs. Azithromycin also is used to treat or prevent disseminated Mycobacterium avium complex (MAC) infection [a type of lung infection that often affects people with human immunodeficiency virus (HIV)]. Azithromycin is in a class of medications called macrolide antibiotics. It works by stopping the growth of bacteria.

  • Antibiotics such as azithromycin will not work for colds, flu, or other viral infections. Using antibiotics when they are not needed increases your risk of getting an infection later that resists antibiotic treatment.

Nilotinib Overview

  • Nilotinib is used to treat certain types of chronic myeloid leukemia (CML; a type of cancer of the white blood cells) who have recently found to have this condition in adults and children 1 year of age and older. It is also used to treat certain types of CML in adults whose disease could not be treated successfully with imatinib (Gleevec) or adults who cannot take imatinib because of side effects. Nilotinib is also used to treat certain types of CML in children 1 year of age or older whose disease could not be treated successfully with other tyrosine kinase inhibitor therapies or who cannot take these medications because of side effects. Nilotinib is in a class of medications called kinase inhibitors. It works by blocking the action of the abnormal protein that signals cancer cells to multiply. This helps to stop or slow the spread of cancer cells.
