- Glucosamine is an amino monosaccharide produced by chondrocyte cells (the cells found in cartilage that are responsible for the synthesis, maintenance, and regeneration of cartilage) and used to make glycosaminoglycans (GAGs) and proteoglycans, the structural molecules that make up the connective tissues of the skin, tendons, ligaments, and joints. By providing the "building blocks" necessary for joint and structural tissue formation, glucosamine supplementation helps to support the maintenance of healthy joint tissues.
Chondroitin sulfate is another very important naturally occurring constituent of cartilage tissue. Chondroitin sulfate has been shown to encourage healthy cartilage production.
MSM (methylsulfonylmethane) is an organic form of the mineral sulfur, found naturally in plants. Sulfur, provided by MSM, promotes healthy skin, joints, bone, hair and nails.

The only side effects reported with chrondroitin sulfate usage appear to be mild gastrointestinal incidents. Similar adverse reactions have been reported with glucosamine and methylsulfonylmethane, yet both of these compounds also are often suggested as supporting GI-tract health and function. It was feared that glucosamine might adversely affect insulin sensitivity, but clinical trials have disproven this. The use of MSM can lead to the occurrence of an abnormal resonance in the in vivo brain proton MR spectrum as well as in the in vitro cerebrospinal fluid MR spectrum, hence should be revealed prior to undergoing these or related diagnostic tests. Vitamin C is well tolerated at the suggested level of intake. Manganese is generally well tolerated. However, supplementation can lead to neuro-toxicity in those with liver failure.
