Use caution when coadministering amodiaquine and deferasirox as this may result in elevated amodiaquine plasma concentrations. Although the interaction between amodiaquine and deferasirox has not been studied, concomitant use of deferasirox (30 mg/kg/day for 4 days), a CYP2C8 inhibitor, and repaglinide (single dose of 0.5 mg), another CYP2C8 substrate, resulted in an increase in repaglinide AUC and Cmax to 2.3-fold and 62%, respectively, in a study of healthy volunteers. Therefore, caution should be used when coadministering deferasirox and amodiaquine, also a CYP2C8 substrate, as this may result in increased amodiaquine plasma concentrations. Monitoring patients for exposure related toxicity is recommended1.
