Interaction Summary
Serotonin syndrome associated with concomitant use of tapentadol and serotonergic drugs have been reported, including SSRIs, serotonin and norepinephrine reuptake inhibitors, tricyclic antidepressants, triptans, 5-HT3 receptor antagonists, drugs that impair serotonin metabolism, and other drugs that affect the serotonergic system; observe the patient especially during treatment initiation and dose adjustment. Symptoms include changes in mental status, tachycardia, hypotension, hyperreflexia, muscle rigidity, nausea, vomiting, and diarrhea; may occur within several hours to a few days of concomitant use but can occur later; discontinue tapentadol if suspected.

How To Manage Interaction
Concomitant use of tapentadol with other serotonergic agents increases the risk of serotonin syndrome. If concomitant use of tapentadol and serotonergic agents is necessary, observe the patient especially during treatment initiation and dose adjustment. Symptoms of serotonin syndrome due to concomitant use generally occur within hours to days of use but can occur later; discontinue tapentadol if suspected.

Venlafaxine is used to treat depression. Venlafaxine extended-release (long-acting) capsules are also used to treat generalized anxiety disorder (GAD; excessive worrying that is difficult to control), social anxiety disorder (extreme fear of interacting with others or performing in front of others that interferes with normal life), and panic disorder (sudden, unexpected attacks of extreme fear and worry about these attacks). Venlafaxine is in a class of medications called selective serotonin and norepinephrine reuptake inhibitors (SNRIs). It works by increasing the amounts of serotonin and norepinephrine, natural substances in the brain that help maintain mental balance.
Tapentadol Overview

Tapentadol tablets are used to treat moderate to severe acute pain (pain that begins suddenly, has a specific cause, and is expected to go away when the cause of the pain is healed). Tapentadol extended-release tablets are used to treat severe neuropathic pain (pain caused by nerve damage) in people who have diabetes. Tapentadol extended-release tablets are only used to treat people who are expected to need medication around-the-clock to relieve pain that cannot be controlled by the use of other pain medications. Tapentadol is in a class of medications called opiate (narcotic) analgesics. It works by changing the way the brain and nervous system respond to pain.
