Benfotiamine is generally well tolerated in short-term studies, though long-term safety hasn't been fully worked out. In one six-week trial of 98 patients taking 300–600 mg daily, a small number reported allergic skin reactions (two participants) and mild stomach discomfort (six participants), at rates no higher than placebo.

Formula

Benfotiamine (S-benzoylthiamine-O-monophosphate) is a derivative of thiamin, belonging to the family of compounds known as "allithiamines." Benfotiamine is fat-soluble and more bioavailable and physiologically active than thiamin. Benfotiamine raises the blood level of thiamin pyrophosphate (TPP), the biologically active co-enzyme of thiamin, and stimulates transketolase, a cellular enzyme essential for maintenance of normal glucose metabolic pathways.
