These amino acids are generally well tolerated at typical doses, though long-term safety data are limited — especially for glycine and L-serine in supplement form. Reported side effects are usually mild: rare sedation, irritability, insomnia, memory problems, headache, and sensory changes with glycine; mild GI upset (soft stools, nausea, vomiting, dry mouth) with glycine; headache and GI upset (diarrhea, constipation, gas, abdominal pain, bloating) with L-threonine; and GI distress (abdominal pain, bloating, nausea, loss of appetite) with L-serine.

Rarely, high doses of L-serine may cause seizures or reversible eye jerking (nystagmus). Pregnancy and breastfeeding data are sparse: L-alanine is likely safe in dietary amounts; L-threonine, glycine, and L-serine have insufficient data, so avoid supplemental doses unless your doctor approves.

Nmda Antagonists

Theoretically, threonine might decrease the effects of NMDA antagonists.
Threonine increases central nervous system (CNS) glycine levels. Glycine seems to bind a site on NMDA receptors and enhance the activity of the receptors.

Clozapine (Clozaril)

Theoretically, glycine might decrease the effectiveness of clozapine.
One small clinical study in patients with schizophrenia shows that adding glycine to clozapine therapy worsens symptoms of schizophrenia when compared with clozapine alone. The mechanism of this interaction is unclear.
