Enzy-Gest contains 10 active ingredients, most of them digestive enzymes that work together to break down food. Amylase breaks down carbohydrates, bromelain and papain digest proteins, lipase breaks down fats, and pepsin and protease are additional protein-digesting enzymes.
Betaine HCl is a stomach-acid enhancer meant to support digestion. The product also includes pancreatin (a natural blend of pancreatic enzymes), ox bile, and pancrelipase—all compounds that aid fat and nutrient absorption in the small intestine.
The tablet also contains several inactive ingredients—fillers and binders like microcrystalline cellulose, stearic acid, magnesium stearate, and others—that give the tablet its form and stability.
Does it work?
Not established
The evidence we hold on the active ingredients in Enzy-Gest shows insufficient reliable evidence to rate their effectiveness for the conditions they're claimed to help. For papain, we have insufficient evidence on cancer, dental cavities, radiation skin damage, muscle soreness, shingles, and intestinal parasites.
Bromelain similarly lacks solid evidence for hay fever, burns, cancer, nerve damage from chemotherapy, circulation problems, and diabetes. Betaine HCl has insufficient evidence for indigestion, low thyroid function, diarrhea, rheumatoid arthritis, food allergies, and hay fever, while lipase has insufficient evidence for inflammatory bowel disease and indigestion.
In short, while these are digestive-support ingredients, the scientific backing for their specific uses is not established in the data we hold.
How safe is it?
Well-documented data
Most of these enzymes are generally well tolerated at normal doses, but there are some important caveats. Papain can irritate the digestive tract and trigger allergic reactions in sensitive people—symptoms may include itchy watery eyes, runny nose, sneezing, stomach cramps, sweating, or diarrhea.
High doses of papain are unsafe. Bromelain commonly causes mild gastrointestinal effects like diarrhea, gas, and stomach upset.
Protease and pepsin may also cause digestive upset or allergic reactions. Betaine HCl is hard on the stomach and is not safe for anyone with ulcers, gastritis, or true acid reflux—it can cause or worsen heartburn.
In pregnancy, the safety data advises against concentrated papain and unripe papaya products. Bromelain safety data is absent, so it's best avoided.
Betaine HCl safety data is also absent, so it should be avoided. For breastfeeding, papain, bromelain, protease, pepsin, and betaine HCl all lack sufficient safety information—talk to your doctor or pharmacist before using this product if you're nursing.

Betaine HCl increases stomach acid and may reduce how well your stomach-acid-blocking medications work—specifically H2-blockers, antacids, and proton pump inhibitors (PPIs). These are Minor interactions, but they do matter if you rely on those medications.

Bromelain
Anticoagulant/Antiplatelet Drugs
Bromelain may have antiplatelet effects and may increase the risk of bleeding if used with anticoagulant or antiplatelet drugs.
There is one case report of a patient experiencing minor bruising while taking bromelain with naproxen. Bromelain is thought to have antiplatelet activity. Whether this interaction is of concern with topical bromelain is unclear. Interference with coagulation of burn wounds has been reported in a patient receiving bromelain-based enzymatic debridement. However, observational research has found that topical bromelain debridement is not associated with increases or decreases in laboratory markers of coagulation when compared with surgical debridement.
Tetracycline Antibiotics
Theoretically, bromelain might increase levels of tetracycline antibiotics.
Laboratory research suggests that bromelain might increase the absorption of tetracycline antibiotics. However, a study in healthy adults reported no difference in tetracycline plasma levels when a 500 mg dose was taken with or without bromelain 80 mg.
Betaine HCl
Antacids
Betaine hydrochloride increases stomach acidity and could decrease the effects of antacids.
In human research, betaine hydrochloride increases stomach acidity. Antacids are taken to decrease stomach acidity. Theoretically, taking betaine hydrochloride along with antacids might decrease the effects of the antacids.
H2-Blockers
Betaine hydrochloride increases stomach acidity and could decrease the effects of H2-blockers.
In human research, betaine hydrochloride increases stomach acidity. H2-blockers are used to decrease stomach acidity. Theoretically, taking betaine hydrochloride along with H2-blockers might decrease the effects of H2-blockers.
Proton Pump Inhibitors (Ppis)
Betaine hydrochloride increases stomach acidity and could decrease the effects of PPIs.
In human research, betaine hydrochloride increases stomach acidity. PPIs are used to decrease stomach acidity. Theoretically, taking betaine hydrochloride along with PPIs might decrease the effects of PPIs
Papain
Warfarin (Coumadin)
Theoretically, papain might increase the effects and side effects of warfarin.
In one case report, a patient previously stable on warfarin was found to have an international normalization ratio (INR) of 7.4, which was attributed to ingestion of a supplement containing papain from papaya extract.
