L-glycine is generally well tolerated at typical doses, but long-term safety data are limited. Rare side effects include mild sedation; irritability, insomnia, fatigue, memory impairment, headache, and sensory impairment have been reported but may not be directly linked to glycine.

Gastrointestinal effects like soft stools, nausea, vomiting, and upper GI discomfort can occur rarely and resolve quickly if you stop the supplement. Papain is generally tolerated as a food but may irritate the digestive tract and cause allergic reactions in sensitive individuals.

High oral doses may be unsafe, and rare serious effects include esophageal perforation and severe gastritis. Bromelain is generally well tolerated short-term, with common side effects including diarrhea, flatulence, gastric upset, and headache; long-term safety data are limited.

Pepsin is generally well tolerated but may cause digestive upset or allergic reactions in some people. Occupational exposure to airborne pepsin or other proteolytic enzymes can trigger allergic rhinitis or asthma.

Anticoagulant/Antiplatelet Drugs

Bromelain may have antiplatelet effects and may increase the risk of bleeding if used with anticoagulant or antiplatelet drugs.
There is one case report of a patient experiencing minor bruising while taking bromelain with naproxen. Bromelain is thought to have antiplatelet activity. Whether this interaction is of concern with topical bromelain is unclear. Interference with coagulation of burn wounds has been reported in a patient receiving bromelain-based enzymatic debridement. However, observational research has found that topical bromelain debridement is not associated with increases or decreases in laboratory markers of coagulation when compared with surgical debridement.

Tetracycline Antibiotics

Theoretically, bromelain might increase levels of tetracycline antibiotics.
Laboratory research suggests that bromelain might increase the absorption of tetracycline antibiotics. However, a study in healthy adults reported no difference in tetracycline plasma levels when a 500 mg dose was taken with or without bromelain 80 mg.

Warfarin (Coumadin)

Theoretically, papain might increase the effects and side effects of warfarin.
In one case report, a patient previously stable on warfarin was found to have an international normalization ratio (INR) of 7.4, which was attributed to ingestion of a supplement containing papain from papaya extract.

Clozapine (Clozaril)

Theoretically, glycine might decrease the effectiveness of clozapine.
One small clinical study in patients with schizophrenia shows that adding glycine to clozapine therapy worsens symptoms of schizophrenia when compared with clozapine alone. The mechanism of this interaction is unclear.
