MacuGuard Ocular Support contains 7 active ingredients. The main ones are lutein, alpha-carotene, trans-zeaxanthin, meso-zeaxanthin, and mixed carotenoids — a group of plant pigments that concentrate in the eye's macula (the part responsible for sharp central vision).
The formula also includes saffron stigma extract and cyanidin-3-glucoside, a compound from plants. These are blended with inactive ingredients like olive oil, gelatin, glycerin, and safflower oil to form the softgel capsule.
Does it work?
Moderate evidence
Lutein and zeaxanthin are rated possibly effective for age-related macular degeneration (AMD) — a common eye condition that can blur central vision as you age. There isn't enough reliable evidence to rate how well lutein or zeaxanthin work for cataracts, though lutein was studied for this.
Saffron extract is rated possibly effective for Alzheimer disease and chemotherapy-induced peripheral neuropathy (nerve pain from cancer treatment), but we hold no effectiveness ratings for this product's use as a general ocular support supplement — that claim rests on the evidence for lutein and zeaxanthin in AMD.
How safe is it?
Well-documented data
Lutein at typical supplement doses is generally well tolerated, with doses up to 20 mg daily showing no adverse effects in trials. Both zeaxanthin forms are also generally well tolerated at standard amounts.
Saffron is mostly well tolerated, but clinical trials reported gastrointestinal complaints, nausea, sedation, and vomiting. Rarely, anaphylaxis (a severe allergic reaction) has occurred — anaphylactic reactions can happen within minutes of eating saffron-prepared food.
There's also a rare report of anxiety and agitation at higher saffron doses. If you have a saffron allergy, avoid this product.

Saffron can add to the sedative effects of CNS depressants (sedating medications and sleep aids), potentially causing extra drowsiness. It may also lower blood pressure when combined with antihypertensive drugs (blood pressure medications), so those on treatment for high blood pressure should monitor closely.
Saffron can theoretically increase the risk of low blood sugar (hypoglycemia) if you take antidiabetes drugs. Additionally, saffron might slow how your body breaks down caffeine, which could raise caffeine levels in your system.
Both zeaxanthin ingredients in this product (trans-zeaxanthin and meso-zeaxanthin) have a Moderate interaction with antidiabetes drugs — theoretically increasing the risk of low blood sugar, though population studies haven't shown this risk with dietary intake. Saffron Stigma Extract
Antidiabetes Drugs
Theoretically, concomitant use of saffron with antidiabetes drugs might increase the risk of hypoglycemia.
Some clinical research shows that taking saffron extract reduces fasting levels of glucose when used in addition to hypoglycemic agents. However, saffron powder itself has not been shown to reduce fasting glucose levels.
Antihypertensive Drugs
Theoretically, concomitant use of saffron with antihypertensive drugs might have additive effects.
Animal and human research suggests that saffron extract can decrease blood pressure.
Caffeine
Theoretically, saffron might inhibit the metabolism of caffeine.
A small clinical study suggests that taking saffron powder 300 mg in 150 mL water daily for 5 days and then taking caffeine 200 mg seems to reduce caffeine metabolite levels in the saliva and urine in males, but not females. Theoretically, this may be due to the inhibition of cytochrome P450 1A2 by saffron.
Cns Depressants
Theoretically, concomitant use of saffron and CNS depressants might have additive sedative effects.
Clinical research shows that taking saffron extract 60 mg orally daily for 26 weeks can cause drowsiness and sedation. Animal research suggests that adding saffron to hexobarbital further increases sleeping and slows motor activity.
Marigold Flower Extract
Cns Depressants
Theoretically, calendula might have additive effects when used with CNS depressants, although this appears to be unlikely.
Although some animal research has suggested that a saponoside constituent in calendula may have sedative effects, calendula has been used for over 30 years without reports of sedation in humans.
