First, let's look at all the drugs in question here:

- guaifenesin (expectorant)
- pseudoephedrine (nasal decongestant)
Delsym (dextromethorphan) is a cough suppressant that works by acting on the "cough center" in the brain. It is best taken for dry unproductive coughs.
If you have a productive cough, meaning you are coughing up phlegm, it is best to avoid Delsym as this could limit your ability to clear the mucus from your lungs (i.e. coughing helps to remove the mucus).
The guaifenesin in Mucinex-D is an expectorant medication that thins and loosens up phlegm or mucus so that you can more easily cough it up clearing it from your throat and lungs. It is most effective if you have a productive cough or the feeling of mucus in your chest.
So, to answer your question, you shouldn't necessarily use both of these medications at the same time, not because it is unsafe, but because it may be counterproductive in treating your symptoms.
Instead, you should use Delsym if you have a dry cough, and Mucinex D if you have a productive cough with sinus or nasal congestion.
Having said all this, the combination of guaifenesin and dextromethorphan can be found in many over-the-counter products (e.g. Robitussin DM).
You may find them effective in improving your symptoms, especially at night (what is more annoying than a nighttime cough!), but I'd suggest trying them separately and see if you don't find that more beneficial.
