Use caution with the concomitant use of brentuximab vedotin (a CYP3A substrate) and this drug, a strong CYP3A inhibitor, due to the potential for significant increases in monomethyl auristatin E (MMAE) plasma concentrations, an active component of brentuximab vedotin. During drug interaction studies, coadministration of brentuximab vedotin and ketoconazole (a strong CY3A4 inhibitor) resulted in a 34% increase in MMAE AUC. As a similar reaction with other strong CYP3A inhibitors cannot be ruled out, monitoring for adverse events is recommended if coadministration is required1.
