Reduced posaconazole exposure and reduced efficacy of posaconazole

Coadministration of posaconazole NOXAFIL® oral suspension and metoclopramide may result in reduced exposure of posaconazole, which may reduce the efficacy of posaconazole. In drug interaction studies, coadministration of posaconazole 400 mg (oral suspension single dose) metoclopramide 10 mg (three times a day for 2 days) resulted in reduced Cmax and AUC of posaconazole by approximately 21% and 19%, respectively. If concomitant use is required, monitor the patient closely for breakthrough fungal infections12.
