a) During a pharmacokinetic drug interaction study, administration of rifAMPin, a potent CYP3A4 inducer, in the presence of romiDEPsin, a CYP3A4 substrate, unexpectedly resulted in an increase in the AUC and Cmax of romiDEPsin by 80% and 60%, respectively, rather than a decrease. Coadministration also decreased the romiDEPsin clearance and volume of distribution by 44% and 52%, respectively. The increase in exposure seen after coadministration with rifAMPin is likely due to rifampin’s inhibition of an undetermined hepatic uptake process that is predominant for the disposition of romiDEPsin 1.
