Concomitant use of suzetrigine with sensitive CYP3A substrates may reduce sensitive CYP3A substrates exposure which may also reduce the efficacy of these substrates. Discontinuation of suzetrigine may increase the exposure of sensitive CYP3A substrates. Dosage adjustment of the concomitant CYP3A substrates may be required when initiating or discontinuing suzetrigine. In a drug interaction study, coadministration of suzetrigine 50 mg every 12 hours and midazolam (sensitive CYP3A substrate), at steady state decreased the geometric mean (90% CI) AUC (0 to inf) of midazolam by 48% (43-53%) and Cmax by 37% (27-45%)1.
