What Severity, Likelihood & Evidence Mean

Severity — How Serious It Can Be

  • Major. Clinically significant; generally best avoided, or used only under direct professional supervision.
  • Moderate. May need monitoring, a dose adjustment, or separating the times you take each one.
  • Minor. Generally not clinically significant, but still worth noting and mentioning to your pharmacist.
  • No known interaction. Checked against our sources with nothing documented — not the same as proven safety.

Likelihood — How Well It’s Documented

  • Likely. Well-controlled human studies have demonstrated the likely existence of this interaction
  • Probable. Interaction has not been documented in well-controlled studies, however, the interaction has been demonstrated in some small human studies or in controlled animal studies in conjunction with multiple case reports.
  • Possible. Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists
  • Unlikely. Interaction has been demonstrated in animal or in lab research but has been shown not to occur in humans.

Where This Data Comes From

  • Interaction records are evidence-graded and sourced from the Natural Medicines database (TRC Healthcare), the same reference used by pharmacists and hospitals.

Trendione: Uses, Safety & Side Effects

The bottom line

Trendione (also marketed as estra-4,9,11-trien-3,17-dione) is a synthetic anabolic steroid sold as a so-called "prohormone" or "designer steroid," not a natural herb or vitamin. It is not a legitimate dietary supplement, has not been proven safe in humans, and is linked to potentially serious side effects, including liver harm. We strongly advise against using it and recommend talking with your doctor or pharmacist.

Overview

Trendione is the common name for a synthetic steroid compound often listed on labels as estra-4,9,11-trien-3,17-dione or "tren-dione." Despite being sold in bottles that look like ordinary supplements, it is not an herb, vitamin, or food. It is a lab-made chemical that is closely related to anabolic steroids.

It has been marketed mainly to bodybuilders and athletes as a "prohormone" or "designer steroid," meaning it is meant to convert into or act like a powerful anabolic steroid once it is in the body. Products like this have appeared in muscle-building and "hardcore" sports supplements.

Because Trendione is a synthetic steroid rather than a true dietary ingredient, it sits in a gray legal and safety area. In many places, steroid-like prohormones have been restricted or pulled from the market.

How it works

Trendione is thought to act as a "prohormone" that the body may convert into an active anabolic steroid, or it may directly stimulate androgen (male hormone) receptors. Anabolic steroids work by telling muscle cells to build more protein, which can increase muscle size and strength.

These proposed mechanisms are based on the chemistry of related steroids and on laboratory reasoning, not on solid human studies of Trendione itself. Activating androgen pathways can also disrupt the body's own hormone balance and stress the liver, which is part of why these compounds are risky.

Safety & precautions

We consider Trendione unsafe for general use. Like other anabolic steroids and prohormones, it has been linked to liver injury, unhealthy changes in cholesterol, and disruption of natural hormone production.

People who should absolutely avoid it include anyone who is pregnant or breastfeeding, teenagers and young adults whose bodies are still developing, and anyone with liver problems, heart disease, high cholesterol, hormone-sensitive conditions, or a history of mood disorders.

Products containing Trendione may also be mislabeled or contain unlisted ingredients, since they are often sold outside normal regulatory oversight. Pregnancy and breastfeeding: avoid completely. If you are using or considering anything like this, please talk with your doctor or pharmacist first.

Side effects

Reported and expected side effects of steroid-like prohormones include liver damage (sometimes serious), acne, oily skin, hair loss, and unhealthy cholesterol changes.

Hormone-related effects can include shrinking of the testicles, reduced sperm count, breast tissue growth in men, and menstrual changes, deepening of the voice, or unwanted hair growth in women. Some users report mood swings, irritability, or aggression.

Rare but serious problems can include liver failure and cardiovascular complications. Any sign of jaundice (yellow skin or eyes), dark urine, severe abdominal pain, chest pain, or unusual swelling should prompt immediate medical attention.

Dosing

There is no established safe or effective dose for Trendione. Because it is a synthetic steroid rather than a studied medicine, no health authority has set a recommended amount.

We do not provide dosing for this product because we do not recommend using it. If you see a product containing Trendione or "estra-4,9,11-trien-3,17-dione," the safest step is to not take it and to speak with a pharmacist or doctor about safer, evidence-based ways to reach your fitness goals.

Pregnancy & Breastfeeding

Trendione & Pregnancy

Possibly Unsafe

When used orally. Prohormones can be converted to active hormones in the body and cause adverse effects such as menstrual irregularities, infertility, hair loss, and acne. Prohormones have also been associated with hepatotoxicity and increased risk of cardiovascular disease. The U.S. Food and Drug Administration and other regulatory agencies warn that dietary supplements containing prohormones are dangerous. Avoid using.

Trendione: Common Questions

What is Trendione used for, and does it work?

See the graded list on this page, and talk to your pharmacist or doctor before relying on it for any condition.
Does Trendione interact with prescription medications?
We do not currently list any known drug interactions for Trendione. This does not guarantee that none exist, so always confirm with your pharmacist before combining it with your medications.
How are Trendione interactions rated?
Each interaction is graded major, moderate, or minor based on how clinically significant it is and the strength of the evidence behind it. Major interactions are the most serious and are best avoided, or used only under close professional supervision.
Is it safe to take Trendione with my medications?
It depends on the specific medication. Some combinations are fine, while others call for monitoring, timing changes, or should be avoided. Check your exact drug above and confirm with your pharmacist or doctor before starting, stopping, or changing anything.

Our interaction data is built on the Natural Medicines database — an evidence-graded reference for vitamins, herbs, and supplements — and is reviewed by licensed HelloPharmacist pharmacists, who may add clinical context.
Is Trendione a natural supplement or herb?
No. Despite being sold in supplement-style bottles, Trendione is a synthetic steroid compound, not a plant, vitamin, or food.
Is Trendione safe to take?
We do not consider it safe. It is a steroid-like prohormone linked to liver damage, hormone disruption, and other serious risks, with no good human safety data.
Can I take Trendione if I'm pregnant or breastfeeding?
No. Anabolic steroids can harm a developing baby or nursing infant, so it should be avoided completely during pregnancy and breastfeeding.
Will Trendione really build muscle?
There are no quality human studies proving it works. Any potential muscle effects come with real risks, and we recommend safer, proven approaches instead.
