Glycine is generally well tolerated at typical doses, but long-term safety data are limited. A small number of trial participants reported mild sedation, though symptoms went away quickly when they stopped taking it.

Others reported irritability, insomnia, fatigue, memory problems, headache, and sensory changes, though it's unclear if glycine actually caused these. Rare gastrointestinal side effects have included soft stools, nausea, vomiting, and upper stomach discomfort — these also resolved quickly with discontinuation.
