Calm contains eight active ingredients. L-Glutamine and L-Glycine are amino acids (building blocks your body uses for various functions) — glutamine supports immune function and gut health, and glycine may have calming effects.
GABA (gamma-aminobutyric acid) is a neurotransmitter that helps regulate nervous system activity. L-Tyrosine is an amino acid your body uses to make neurotransmitters that support mood and focus.
Inositol is a carbohydrate-like compound that plays a role in cell signaling. DMAE (dimethylaminoethanol) is thought to support brain function.
Motherwort is a plant traditionally used for relaxation and heart health. Wintergreen is included for its properties, though it's present in small amounts in a capsule product.
The remaining ingredients are inactive — vegetarian capsule material, rice flour, microcrystalline cellulose, silica, and ascorbyl palmitate (a form of vitamin C) — which serve as fillers, binders, and preservatives.
Does it work?
Moderate evidence
The evidence is mixed and often limited for most uses in this product. L-Tyrosine is possibly effective for cognitive function and memory, and it's effective for phenylketonuria (PKU), a genetic condition.
Inositol is possibly effective for polycystic ovary syndrome (PCOS) and metabolic syndrome. L-Glycine may be possibly effective for schizophrenia.
For the other ingredients and most other claimed uses — anxiety, sleep, stress relief, or athletic performance — the evidence we hold rates as insufficient to establish effectiveness, or in some cases (like DMAE for Alzheimer's disease, or inositol for anxiety) the evidence suggests they may not work. The product hasn't been tested for its specific combination in any condition.
How safe is it?
Well-documented data
Most of these ingredients are generally well tolerated at common doses, though side effects do occur. L-Glutamine, L-Tyrosine, GABA, and Inositol can cause digestive upset — nausea, diarrhea, bloating, or stomach discomfort.
L-Glycine is generally well tolerated but rarely causes mild sedation. DMAE can cause abdominal cramps, diarrhea, nausea, or vomiting, and at higher doses (over 1,000 mg daily) has been linked to mood changes.
Motherwort may cause diarrhea or stomach irritation. Wintergreen leaf appears safe orally, but wintergreen oil is toxic if swallowed.
For pregnancy: L-Glutamine is rated likely safe; Inositol is rated possibly safe in pregnancy but should be used only under medical supervision; Motherwort is rated likely unsafe and should be avoided; and L-Tyrosine, L-Glycine, and DMAE have insufficient data — talk to your doctor or pharmacist before use. For breastfeeding: L-Glutamine, L-Tyrosine, L-Glycine, and DMAE have insufficient safety data — use only if your clinician advises it.
GABA and Motherwort are rated unsafe while breastfeeding and should be avoided. Inositol has no lactation data on file.

Also check if you're on anticonvulsants (seizure drugs), blood pressure medications, antihypertensive drugs, CNS depressants (sleep aids or sedatives), levodopa (Parkinson's), thyroid hormone, antidiabetes drugs, clozapine (an antipsychotic), anticholinergic drugs, or cholinergic drugs.

The most serious interaction we've documented is wintergreen with warfarin (a blood thinner). Wintergreen oil can be absorbed through the skin and increase warfarin's effects, raising your bleeding risk — case reports show this can happen with topical products.

Glutamine may theoretically work against anticonvulsant medications (drugs that prevent seizures), though this hasn't been reported in humans yet. GABA might lower blood pressure when combined with antihypertensive drugs and could add to the sedative effects of CNS depressants (sleep aids, sedatives, some pain relievers).
L-Tyrosine can interfere with levodopa (used for Parkinson's disease) and might amplify thyroid hormone medications. L-Glycine may reduce how well clozapine (an antipsychotic) works.
Inositol could increase the risk of low blood sugar (hypoglycemia) if you're on diabetes medications. DMAE might decrease the effects of anticholinergic drugs or intensify cholinergic drugs.
Motherwort may add to the sedative effects of CNS depressants. Wintergreen also poses a moderate risk with aspirin due to shared salicylate content.

GABA
Antihypertensive Drugs
Theoretically, taking GABA with antihypertensive drugs might increase the risk of hypotension.
Some clinical research shows that GABA can decrease blood pressure in patients with hypertension.
Cns Depressants
Theoretically, GABA might have additive sedative effects when used in conjunction with CNS depressants. However, it is unclear if this concern is clinically relevant.
Endogenous GABA has well-established relaxant effects and GABA(A) receptors have an established physiological role in sleep. However, the effects of GABA supplements are unclear, as it is unknown whether exogenous GABA crosses the blood-brain barrier. Although there have been limited reports of drowsiness or tiredness with GABA supplements, these effects have not been widely reported in clinical studies. Additionally, intravenous GABA 0.1-1 mg/kg has been shown to induce anxiety in a dose-dependent manner.
Motherwort
Cns Depressants
Theoretically, taking motherwort concomitantly with other CNS depressants may increase the risk of sedation.
Motherwort has been reported to cause CNS depression and sedation.
DMAE
Anticholinergic Drugs
Theoretically, deanol might decrease the effectiveness of anticholinergic drugs.
Deanol is thought to increase acetylcholine levels.
Cholinergic Drugs
Theoretically, deanol might increase the effects and adverse effects of cholinergic drugs.
Deanol is thought to increase acetylcholine levels.
Inositol
Antidiabetes Drugs
Theoretically, taking inositol with antidiabetes drugs might increase the risk of hypoglycemia.
Clinical research shows that inositol lowers blood glucose levels and glycated hemoglobin (HbA1c) levels in patients with diabetes.
L-Glutamine
Anticonvulsants
Theoretically, glutamine might antagonize the effects of anticonvulsant medications.
Glutamine is metabolized to the excitatory neurotransmitter glutamate. Glutamate might have antagonistic effects with anticonvulsant drugs. However, this interaction has not yet been reported in humans.
Wintergreen
Warfarin (Coumadin)
Using wintergreen oil topically might increase the effects and adverse effects of warfarin.
Several case reports show that concomitant use of topical wintergreen oil-containing products and warfarin can increase INR and bleeding risk due to systemic absorption of the methyl salicylate contained in wintergreen oil. Topical analgesic gels, lotions, creams, ointments, liniments, and sprays can contain up to 55% methyl salicylate.
Aspirin
Using wintergreen oil topically in large amounts, with occlusive dressings, or for prolonged periods of time might increase the risk of additive salicylate toxicity when used with aspirin.
Wintergreen oil contains methyl salicylate. Topical analgesic gels, lotions, creams, ointments, liniments, and sprays can contain up to 55% methyl salicylate.
L-Tyrosine
Levodopa
Theoretically, tyrosine might decrease the effectiveness of levodopa.
Tyrosine and levodopa compete for absorption in the proximal duodenum by the large neutral amino acid (LNAA) transport system. Advise patients to separate doses of tyrosine and levodopa by at least 2 hours.
Thyroid Hormone
Theoretically, tyrosine might have additive effects with thyroid hormone medications.
Tyrosine is a precursor to thyroxine and might increase levels of thyroid hormones.
L-Glycine
Clozapine (Clozaril)
Theoretically, glycine might decrease the effectiveness of clozapine.
One small clinical study in patients with schizophrenia shows that adding glycine to clozapine therapy worsens symptoms of schizophrenia when compared with clozapine alone. The mechanism of this interaction is unclear.
