Lactase is generally well tolerated. Lipase is also generally well tolerated in supplement form, though people with serious digestive conditions should only use it under medical guidance.

Both have limited safety data in pregnancy and breastfeeding — the facts advise checking with your doctor before regular use during pregnancy and before using while nursing. One case of allergic reaction to powdered lactase has been reported (contact dermatitis and allergic rhinoconjunctivitis in a worker exposed to it); this is very rare.

Theoretically, papain might increase the effects and side effects of warfarin.
In one case report, a patient previously stable on warfarin was found to have an international normalization ratio (INR) of 7.4, which was attributed to ingestion of a supplement containing papain from papaya extract.
