Our pharmacy team’s full take, with four database checks built into the cards below — a summary of what is known, not a grade of the product itself.
What’s inside
Full disclosure
This product contains one active ingredient: 500 mg of L-tryptophan, an amino acid your body uses to make serotonin and melatonin — neurotransmitters that influence mood, sleep, and relaxation. The capsule also contains three inactive ingredients (hypromellose, microcrystalline cellulose, and calcium laurate) used to form and hold the capsule together.
Does it work?
Not established
The evidence for L-tryptophan's effectiveness is mixed and limited. For depression, the data rates it possibly ineffective.
For anxiety, athletic performance, teeth grinding (bruxism), and premenstrual dysphoric disorder, there isn't enough reliable evidence to know whether it works. If you're considering it for one of these uses, talk with your doctor or pharmacist about whether the current evidence supports it for your situation.
How safe is it?
Well-documented data
L-tryptophan is generally well tolerated when taken by mouth. The most common side effects are mild: belching, diarrhea, drowsiness, dry mouth, flatulence, headache, heartburn, lightheadedness, nausea, stomach pain, blurred vision, and vomiting.
A serious rare side effect is eosinophilia-myalgia syndrome (EMS), a neurological disorder — but nearly all documented cases in the U.S. were traced to contaminated L-tryptophan from a single Japanese manufacturer in 1989 and led to an FDA recall. Modern products are monitored to prevent that contamination.
For pregnancy, safety data shows mixed ratings: some evidence suggests it may be possibly unsafe, so talk with your doctor before using it if you're pregnant. For breastfeeding, safety isn't established, so it's best avoided unless your healthcare provider advises otherwise.
Meds to double-check
Major interaction found
Before taking L-Tryptophan 500 mg, double-check with your doctor or pharmacist if you take any CNS depressants (sleep aids, sedatives, opioids, alcohol) — the risk is Major for additive drowsiness. Also confirm if you're on serotonergic drugs like antidepressants or certain migraine medications — there's a Moderate risk of boosting serotonin too much.
The bottom line
Scorecard at a glanceFully disclosed formula with no established evidence rating for its marketed use. Major medication interactions have been identified, and safety information is well characterized.
L-tryptophan may appeal to people interested in supporting mood and sleep naturally, but the evidence for depression and other conditions remains weak. If you take any CNS depressant (like sleep aids or opioids) or a serotonergic drug (like an antidepressant), you need to check with your doctor or pharmacist before starting this supplement — the interaction risks are real and potentially serious.
Pregnant or breastfeeding? Talk it over with your healthcare provider first.

L-Tryptophan 500 mg by Doctor's Best Drug Interactions
HelloPharmacist Interaction Report
Doctor's Best L-Tryptophan 500 mg interacts with two major drug categories.
The most serious concern is with CNS depressants (sedatives, sleep aids, opioids, alcohol) — L-tryptophan can add to their drowsy and sedative effects, which the data shows it causes on its own.
Read the full breakdown — every affected drug type, severity by severity
It also interacts with serotonergic drugs (antidepressants, certain migraine and pain medications). Because L-tryptophan is a building block for serotonin, combining it with these drugs might increase serotonin in your system beyond what's intended, raising the risk of serotonin syndrome — a potentially serious condition that includes agitation, confusion, muscle stiffness, and rapid heart rate.
Although the facts mention theoretically more drugs may be affected, interaction data for other medications on file remain limited. Altogether, these interactions span 394 individual medications.
