What’s inside
Full disclosure
This product contains one active ingredient: L-tryptophan, an amino acid your body uses to make serotonin and other brain chemicals. The capsule also holds inactive ingredients — hypromellose, rice flour, ascorbyl palmitate, and stearic acid — which help form and preserve the supplement.
Does it work?
Not established
The evidence we hold shows that L-tryptophan is possibly ineffective for depression. For anxiety, athletic performance, teeth grinding (bruxism), and premenstrual dysphoric disorder (PMDD), we don't have enough reliable evidence to say whether it works or not.
How safe is it?
Well-documented data
L-tryptophan is generally well tolerated, but side effects do occur. The most common are belching, diarrhea, drowsiness, dry mouth, flatulence, headache, heartburn, lightheadedness, nausea, stomach pain, blurred vision, and vomiting.
Serious side effects are rare, but L-tryptophan has a past history tied to a contaminated product: in 1989, over 1,500 people in the US developed a rare neurological disorder called eosinophilia-myalgia syndrome (EMS). Nearly all cases came from L-tryptophan made by one Japanese manufacturer.
The FDA restricted L-tryptophan imports afterward, and modern products are much more carefully regulated, but the history warrants caution. There is not enough safety data for use during pregnancy, so the safety data advises against it.
Safety is also not established for breastfeeding, so avoid use while nursing.
Meds to double-check
Major interaction found
Before taking this product, double-check with us if you're on any CNS depressants (sleeping pills, sedatives, opioids, alcohol) — the drowsiness risk is Major. Also flag any serotonergic drugs (antidepressants, certain migraine or pain medications) — the serotonin syndrome risk is Moderate.
Use the medication checker on this page with your exact prescriptions.
The bottom line
Scorecard at a glanceFully disclosed formula with no established evidence rating for its marketed use. Major medication interactions have been identified, and safety information is well characterized.
L-tryptophan might be worth considering if you're curious about mood support, but the evidence for depression is weak. If you take any sedatives, antidepressants, migraine drugs, or pain medications, you must check with us first — the interaction risks are real.

L-Tryptophan 500 mg by NOW Drug Interactions
HelloPharmacist Interaction Report
L-Tryptophan 500 mg by NOW does interact with certain medications.
The most serious concern is CNS depressants — drugs that slow your brain and nervous system, like sedatives, sleeping pills, opioids, and alcohol. Taking L-tryptophan with these can stack their drowsy effects on top of each other, and we know from research that L-tryptophan itself causes fatigue and drowsiness.
Read the full breakdown — every affected drug type, severity by severity
The second major interaction involves serotonergic drugs — medications that work through serotonin, a brain chemical. This includes antidepressants like SSRIs, SNRIs, and tricyclics, as well as some migraine and pain drugs.
L-tryptophan is a building block your body uses to make serotonin, so combining it with these drugs might boost serotonin too much, raising the risk of serotonin syndrome (a serious condition with agitation, confusion, rapid heartbeat, and muscle rigidity) and changes in blood vessel tone.
Additionally, check your exact medications below before you start — we need to make sure nothing you're taking falls into either category.
