Taken by mouth, L-Tryptophan is generally well tolerated. Common side effects include belching, diarrhea, drowsiness, dry mouth, gas, headache, lightheadedness, nausea, stomach pain, blurred vision, and vomiting.

The supplement has a serious historical caveat: in 1989, contaminated L-Tryptophan from a single Japanese manufacturer was linked to over 1,500 cases of eosinophilia-myalgia syndrome (EMS), a rare neurological disorder involving muscle pain, weakness, and skin changes. Almost all cases traced back to that one source, and the product was recalled in the U.S. with importation limits now in place.

Regarding pregnancy and breastfeeding, the safety data are inconsistent — some evidence suggests it may be possibly unsafe in pregnancy, while other data say it's likely safe, so there isn't enough information to be certain either way.
