Tri-Chromium 500 mcg has 4 active ingredients: three different forms of chromium (Chromium Picolinate, Albion Chromium Nicotinate Glycinate, and ChromeMate, also called Chromium Polynicotinate) and Cinnamon. For chromium deficiency, the data shows chromium is likely effective — it addresses the actual shortage. For type 2 diabetes, the evidence suggests it's possibly effective: some research points to chromium lowering blood glucose, especially in people whose diabetes isn't well controlled.
For prediabetes, schizophrenia, and high blood pressure, the evidence is insufficient or shows it's possibly ineffective. Cinnamon's effectiveness for any of its traditional uses — amenorrhea, angina, cold symptoms, diabetes, diarrhea, or cholesterol — is not established in our data; there isn't enough reliable evidence to rate it either way.
How safe is it?
Well-documented data
Chromium is generally well tolerated at typical supplement doses, but high or long-term doses may pose risks. The most common side effects are stomach upset, headaches, insomnia, irritability, and mood changes.
Rare but serious effects include kidney or liver damage, a muscle breakdown condition (rhabdomyolysis), and low platelet counts (thrombocytopenia). Skin rashes and contact dermatitis have also been reported, especially in people with a chromium allergy.
Cinnamon at food amounts is likely safe, but supplement doses—especially high ones—carry concern about liver toxicity from a compound called coumarin if used long-term. One person taking cinnamon reported a hypoglycemic seizure, though it's unclear if cinnamon caused it.
For pregnancy, chromium is rated likely safe or possibly safe, and cinnamon is likely safe; however, extra supplement doses of chromium should be avoided unless your doctor recommends it, and high-dose cinnamon should be avoided in pregnancy due to limited safety data.

The most serious concern is with diabetes drugs (antidiabetes agents and insulin): chromium may lower blood sugar on its own, and taking it together with these medications could add up and raise your risk of low blood sugar (hypoglycemia). If you take insulin or a diabetes pill, you'll want to check this with your pharmacist or doctor first.

Chromium also interacts with levothyroxine (Synthroid), a thyroid hormone replacement. Research shows chromium can bind to levothyroxine in your digestive tract and reduce how much your body absorbs — one study found a 17% drop in levothyroxine levels.
If you take thyroid medication, space out this supplement and your levothyroxine dose by several hours. Cinnamon adds another theoretical concern: large doses may have additive effects with diabetes drugs and potentially with medications that stress the liver (hepatotoxic drugs).
Two more minor interactions came up: nonsteroidal anti-inflammatory drugs (NSAIDs) like ibuprofen and naproxen may increase chromium levels in your body, and aspirin may boost chromium absorption. Altogether, these interactions span 506 individual medications.

Cinnamon
Antidiabetes Drugs
Theoretically, cassia cinnamon may have additive effects with antidiabetes drugs.
Cassia cinnamon may lower blood glucose levels, and have additive effects in patients treated with antidiabetic agents. Dose adjustments to diabetes medications might be necessary.
Hepatotoxic Drugs
Theoretically, large doses of cassia cinnamon might cause additive effects when used with hepatotoxic drugs.
There is some concern that ingesting large amounts of cassia cinnamon for an extended duration might cause hepatotoxicity in some people. Cassia cinnamon contains coumarin, which can cause hepatotoxicity in animal models. In humans, very high doses of coumarin from 50-7000 mg/day can result in hepatotoxicity that resolves when coumarin use is discontinued. Lower amounts might also cause liver problems in sensitive people, such as those with liver disease or those taking potentially hepatotoxic agents.
Chromium
Antidiabetes Drugs
Theoretically, chromium may have additive effects with antidiabetic agents and increase the risk of hypoglycemia.
Some research shows that taking chromium might lower blood glucose levels, especially in patients with poorly controlled type 2 diabetes.
Insulin
Theoretically, concomitant use of chromium and insulin might increase the risk of hypoglycemia.
In clinical research, chromium has been shown to increase insulin sensitivity,
Levothyroxine (Synthroid, Others)
Chromium might bind levothyroxine in the intestinal tract and decrease levothyroxine absorption.
Clinical research in healthy volunteers shows that taking chromium picolinate 1000 mcg with levothyroxine 1 mg decreases serum levels of levothyroxine by 17% when compared to taking levothyroxine alone. Advise patients to take levothyroxine at least 30 minutes before or 3-4 hours after taking chromium.
Aspirin
Theoretically, aspirin might increase chromium absorption.
Animal research suggests that aspirin may increase chromium absorption and chromium levels in the blood.
Nonsteroidal Anti-Inflammatory Drugs (Nsaids)
NSAIDs might increase chromium levels in the body.
Drugs that are prostaglandin inhibitors, such as NSAIDs, seem to increase chromium absorption and retention.
