Concomitant use of brigatinib (CYP3A substrate) and a moderate CYP3A inhibitor may increase the exposure of brigatinib and increase the incidence of adverse events. A moderate CYP3A inhibitor is predicted to increase the AUC of brigatinib by approximately 40% and therefore should be avoided. If use is unavoidable, reduce the once daily brigatinib dose by approximately 40% (ie, from 180 mg to 120 mg, 120 mg to 90 mg, or from 90 mg to 60 mg). Following discontinuation of the moderate CYP3A inhibitor, resume the brigatinib dose that was tolerated prior to the initiation of the moderate CYP3A inhibitor1.
