Caution is advised when using brentuximab vedotin, a CYP3A4 substrate, together with fluconazole, a moderate CYP3A4 inhibitor. Concomitant use may result in increased plasma concentrations of MMAE1. Although not studied specifically with fluconazole, concomitant use with ketoconazole (a potent CYP3A4 inhibitor) increased MMAE exposure by approximately 34% 2. Fluconazole-mediated CYP3A4 inhibition may persist for 4 to 5 days after discontinuation.1. If concomitant use necessary, consider monitoring for brentuximab vedotin-associated toxicities including neutropenia.
