Concomitant use of ponatinib and agents that elevate gastric pH may decrease ponatinib efficacy. The solubility of ponatinib is pH-dependent. Relative to administration of ponatinib alone, coadministration of a single dose of ponatinib 45 mg in the presence of lansoprazole 60 mg daily in healthy volunteers (n=18) decreased ponatinib AUC by 6% and Cmax by 25%. Despite this interaction, the change in ponatinib exposure is minimal and ponatinib may still be administered concomitantly with these agents1. Consider monitoring for lack of response to ponatinib.
