Ponatinib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Acacia Rigidula interacts with.

Theoretically, Acacia rigidula products that are adulterated with beta-methylphenylethylamine (BMPEA) might increase the levels of CYP3A4 substrates.
In vitro research shows that BMPEA, an adulterant found in many Acacia rigidula products, inhibits CYP3A4 enzyme by 37%.

Here's the concern with pairing Acacia rigidula products with ponatinib (Iclusig). Your body clears ponatinib using a liver enzyme called CYP3A4. Some Acacia rigidula supplements have been found spiked with a stimulant called BMPEA, and lab studies suggest BMPEA can partly block that same enzyme (by roughly a third in a test tube). If that happens in the body, ponatinib could clear more slowly and build up, which may mean stronger effects and more side effects.

With ponatinib that matters, because it can already raise the risk of blood clots, high blood pressure, heart problems, and liver issues. Keep in mind this is theoretical, based on lab research only, not proven in people. Still, given ponatinib's risks, please don't add this supplement without checking with your oncologist or pharmacist first.

Acacia rigidula is a shrub from Texas and Mexico that is marketed in weight-loss and energy supplements. There is very little reliable human evidence that it works, and some products have been found to contain an unapproved stimulant drug (BMPEA), which raises real safety concerns. About Ponatinib

What Is Ponatinib Used For?

Brand name: Iclusig

Ponatinib, sold as Iclusig, treats two types of blood cancer in adults. For Philadelphia chromosome-positive acute lymphoblastic leukemia (Ph+ ALL), Iclusig is used in combination with chemotherapy for newly diagnosed patients, or as a single agent for patients who cannot take other kinase inhibitors or who have a specific mutation called T315I. For chronic myeloid leukemia (CML), Iclusig treats chronic phase CML when at least two prior kinase inhibitors have not worked or caused intolerable side effects, accelerated or blast phase CML when no other kinase inhibitors are suitable, and any phase of CML with the T315I mutation.

Iclusig is not approved or recommended for newly diagnosed chronic phase CML.
