Interaction Deep Dive
Avoid concomitant administration of belinostat with UGT1A1 inhibitors, such as probenecid. Belinostat is primarily metabolized by UGT1A1. Concomitant use with a UGT1A1 inhibitor may increase belinostat exposure, which may increase the risk of belinostat-related adverse reactions. If concomitant use of probenecid is unavoidable, reduce the belinostat dosage by 25%. Belinostat starting dosage of 1,000 mg/m2 to a modified dosage of 750 mg/m2. Belinostat starting dosage of 750 mg/m2 to a modified dosage of 562.5 mg/m2. Belinostat starting dosage of 500 mg/m2: Interrupt belinostat treatment for the duration of probenecid use. After discontinuation of probenecid of 5 half-lives, resume the belinostat dosage that was taken prior to initiating probenecid1.
