Moderate evidence
The evidence for these ingredients is mixed. Beta-carotene is effective for vitamin A deficiency and possibly effective for oral leukoplakia, aging skin, measles, ulcerative colitis, and bronchopulmonary dysplasia.
Lutein is possibly effective for age-related macular degeneration and cataracts. Canthaxanthin is possibly effective for erythropoietic protoporphyria.
For astaxanthin, the evidence we hold is insufficient to rate it for age-related cognitive decline, macular degeneration, aging skin, Alzheimer disease, athletic performance, or carpal tunnel syndrome—meaning the science hasn't yet established whether it works for these uses.
How safe is it?
Well-documented data
Beta-carotene is generally well tolerated at recommended doses, but high doses build up in your body and can become toxic, causing fatigue, dry skin, hair loss, and in severe cases liver damage or pseudotumor cerebri. Astaxanthin is generally well tolerated in studies, though abdominal pain, diarrhea, and red-colored stools have been reported; long-term safety data are limited.
Lutein from food and typical supplements is generally safe; doses up to 20 mg daily have not caused adverse effects. Canthaxanthin at high doses can cause orange-brown skin discoloration, eye deposits with reduced vision, diarrhea, nausea, and in rare cases serious blood disorders.

The most serious concern is beta-carotene (a form of vitamin A), which carries Major severity interactions with retinoids—vitamin A derivatives used in acne and anti-aging medications. Taking retinoids alongside this product could cause dangerously high vitamin A levels and additive toxic effects.

Beta-carotene also has Moderate interactions with blood thinners like warfarin (where high doses may increase bleeding risk), certain antibiotics called tetracyclines (which combined with high-dose vitamin A may raise the risk of pseudotumor cerebri—a serious swelling of brain tissue), and hepatotoxic drugs (potentially liver-damaging medications, where the combination could worsen liver risk). Astaxanthin itself carries Moderate interactions with drugs broken down by your liver's CYP3A4 and CYP2B6 enzymes—theoretically it may lower their levels, though this hasn't been proven in humans yet.
Lutein and canthaxanthin were checked and show no documented interactions in our data. Altogether, these interactions span 903 individual medications.

Astaxanthin
Cytochrome P450 2B6 (Cyp2B6) Substrates
Theoretically, astaxanthin may decrease levels of drugs metabolized by CYP2B6.
In vitro research shows that astaxanthin induces cytochrome CYP2B6 enzyme activity in human hepatocytes. This effect has not been reported in humans.
Cytochrome P450 3A4 (Cyp3A4) Substrates
Theoretically, astaxanthin may decrease levels of drugs metabolized by CYP3A4.
In vitro research shows that astaxanthin induces CYP3A4 enzyme activity in human hepatocytes. This effect has not been reported in humans.
Beta-Carotene
Retinoids
Concomitant use of retinoids with vitamin A supplements might produce supratherapeutic vitamin A levels.
Retinoids, which are vitamin A derivatives, could have additive toxic effects when taken with vitamin A supplements.
Hepatotoxic Drugs
Theoretically, taking high doses of vitamin A in combination with other potentially hepatotoxic drugs might increase the risk of liver disease.
The tolerable upper intake level (UL) is the highest level of intake that is likely to pose no risk of adverse effects. Doses of vitamin A above the UL can cause hepatotoxicity, ranging from elevated liver enzymes to liver failure.
Tetracycline Antibiotics
Theoretically, taking tetracycline antibiotics with high doses of vitamin A can increase the risk of pseudotumor cerebri.
Benign intracranial hypertension (pseudotumor cerebri) can occur with tetracyclines and with acute or chronic vitamin A toxicity. Case reports suggest that taking tetracyclines and vitamin A concurrently can increase the risk of this condition. Avoid high doses of vitamin A in people taking tetracyclines chronically.
Warfarin (Coumadin)
Theoretically, high doses of vitamin A could increase the risk of bleeding with warfarin.
Vitamin A toxicity is associated with hemorrhage and hypoprothrombinemia, possibly due to vitamin K antagonism. Advise patients taking warfarin to avoid doses of vitamin A above the tolerable upper intake level of 10,000 IU/day for adults.
