What’s inside
Full disclosure
This product contains one active ingredient: L-Tryptophan 1000 mg per tablet. L-Tryptophan is an amino acid—a building block your body uses to make proteins and other compounds.
It's also a precursor to serotonin, a brain chemical involved in mood, sleep, and other functions. The remaining ingredients are inactive fillers and binders that help form the tablet.
Does it work?
Not established
The evidence we hold shows L-Tryptophan is possibly ineffective for depression. For anxiety, athletic performance, teeth grinding (bruxism), and premenstrual dysphoric disorder (PMDD), there isn't enough reliable evidence to say one way or the other whether it works.
How safe is it?
Well-documented data
L-Tryptophan is generally well tolerated by mouth, but it can cause common side effects including drowsiness, nausea, diarrhea, headache, dry mouth, stomach pain, heartburn, flatulence, belching, lightheadedness, visual blurring, and vomiting. Rarely, it has been linked to a serious neurological condition called eosinophilia-myalgia syndrome (EMS)—though almost all documented cases in the U.S. came from contaminated product made by a single Japanese manufacturer in 1989, leading to an FDA recall and import restrictions.
There isn't enough safety data to recommend this during pregnancy, so it's best avoided then. Safety is not established for breastfeeding, so you should avoid it while nursing.
If you're pregnant, breastfeeding, or planning to be, talk with your pharmacist or doctor before taking this supplement.
Meds to double-check
Major interaction found
Before taking this supplement, check your medications against these drug types using the interaction tool on this page: CNS depressants (Major severity)—sedatives, sleep aids, alcohol, opioid pain relievers—and serotonergic drugs like antidepressants and certain migraine or pain medications (Moderate severity). No interactions are documented for other medication types.
The bottom line
Scorecard at a glanceFully disclosed formula with no established evidence rating for its marketed use. Major medication interactions have been identified, and safety information is well characterized.
L-Tryptophan may be worth considering if you're interested in mood or sleep support and your medications don't include CNS depressants or serotonergic drugs—but the evidence for many uses is thin. If you take any sedatives, sleep aids, antidepressants, pain or migraine medications, or anything that affects serotonin, check your specific drugs first.

Double Strength L-Tryptophan 1000 mg by NOW Drug Interactions
HelloPharmacist Interaction Report
NOW's Double Strength L-Tryptophan contains a single active ingredient that does interact with medications.
The most serious concern is with CNS depressants—drugs that slow your brain and nervous system, like sedatives, sleep aids, alcohol, and opioid pain relievers. L-Tryptophan can add to their drowsy or sedating effects, and clinical research shows it causes fatigue and drowsiness on its own.
Read the full breakdown — every affected drug type, severity by severity
You'll also want to be careful combining this with serotonergic drugs (medications that boost serotonin), which include many antidepressants and some migraine or pain medications. Adding L-Tryptophan to these raises the theoretical risk of serotonin-related side effects, including serotonin syndrome—a potentially serious condition where serotonin levels climb too high—and problems with blood vessel constriction in the brain.
Altogether, these interactions span 394 individual medications. Before starting this supplement, run your exact medications through the interaction checker on this page.
