Our pharmacy team’s full take, with four database checks built into the cards below — a summary of what is known, not a grade of the product itself.
What’s inside
Full disclosure
This blend contains 4 active ingredients. Chromium polynicotinate is a form of chromium, a mineral that plays a role in how your body handles blood sugar.
Raspberry ketone is a compound isolated from raspberries. Forskolin comes from Coleus forskohlii root extract and is the plant's active component.
An Enzyme Blend rounds out the formula — we don't have specific details on which enzymes or their amounts. The capsule also contains two inactive ingredients: cellulose (a plant-based filler) and diatomaceous earth (a naturally derived additive).
Does it work?
Moderate evidence
Chromium is likely effective for treating actual chromium deficiency, and possibly effective for diabetes — some research suggests it may help lower blood sugar in people with poorly controlled type 2 diabetes. For prediabetes and several other conditions the blend might target, chromium rates as possibly ineffective.
Raspberry ketone's effectiveness for weight management and hair loss is not established — the evidence we hold rates it as having insufficient reliable data. We cannot assess Forskolin or the Enzyme Blend because effectiveness data isn't on file for them.
How safe is it?
Well-documented data
Chromium is generally well tolerated at typical supplement doses, though long-term or high doses carry unknown risks. The most common side effects are gastrointestinal irritation, headaches, insomnia, irritability, and mood changes.
Serious adverse effects are rare but include kidney and liver damage and blood disorders. Skin reactions (rashes, hives) have also been reported.
Raspberry ketone has very limited human safety data at high supplement doses. Because it resembles a stimulant compound, heart palpitations, elevated blood pressure, and abnormal heart rhythms have been reported in case reports.
Two cases describe sweating, diarrhea, and shakiness. Chromium is not recommended in pregnancy beyond what's in a prenatal vitamin unless your doctor advises it.
Raspberry ketone should be avoided in pregnancy and breastfeeding due to insufficient safety information.
Meds to double-check
Major interaction found
Before taking this product, double-check with your pharmacist if you take stimulant drugs (decongestants, ADHD medications, or certain weight-loss products) — raspberry ketone may increase your risk of heart palpitations and dangerous arrhythmias. Also check if you take antidiabetes medications or insulin, because chromium may lower blood sugar too much.
If you take levothyroxine (Synthroid or generics) for thyroid disease, chromium may reduce how much you absorb — your dose might need adjustment. Finally, if you take warfarin (Coumadin), raspberry ketone may require a warfarin dose increase to keep your blood thin enough.
The bottom line
Scorecard at a glanceFully disclosed formula with some supporting evidence for its stated purpose. Major medication interactions have been identified, and safety information is well characterized.
If you take diabetes or insulin medication, thyroid medication, warfarin, or any stimulant drug, you need to check this product with your pharmacist or doctor first — the interactions are real and could affect how your medications work or increase side effects. Otherwise, chromium is generally well tolerated at normal doses, but the safety data on the other ingredients is limited.
Talk to your pharmacist if you're pregnant or breastfeeding, or if you have heart, blood pressure, or liver concerns.

Forskolin Lean Blend by Signature Supplements Drug Interactions
HelloPharmacist Interaction Report
Signature Supplements Forskolin Lean Blend contains ingredients that do interact with medications.
The most serious concern is raspberry ketone, which may increase the risk of dangerous heart rhythm problems and other cardiovascular effects when taken with stimulant drugs (medications like decongestants, some ADHD drugs, and certain weight-loss products). This is a Moderate severity interaction.
Read the full breakdown — every affected drug type, severity by severity
Chromium polynicotinate, another active ingredient here, has Moderate interactions with diabetes and insulin medications — it may lower blood sugar on its own, which could combine with these drugs to push your glucose too low. It also may decrease how much of your thyroid medication (levothyroxine/Synthroid) your body absorbs; research shows a potential 17% drop in thyroid hormone levels.
Additionally, raspberry ketone may interfere with warfarin (a blood thinner), requiring a dose adjustment to keep it working properly. Chromium has Minor interactions with NSAIDs and aspirin, which can raise chromium levels in your body.
We cannot check two of the four ingredients — Enzyme Blend and Forskolin — because we hold no interaction data for them. Altogether, these interactions span 335 individual medications.
Please run your exact medications through the interaction checker on this page before you start.

Forskolin, a constituent of coleus, and nitrates both cause coronary vasodilatory effects.
Anticoagulant/Antiplatelet Drugs
Theoretically, concomitant use of coleus and anticoagulant or antiplatelet drugs might increase the risk of bruising and bleeding.
In vitro and animal research shows that forskolin, a constituent of coleus, can inhibit platelet aggregation and adhesion.
Antihypertensive Drugs
Theoretically, combining coleus with antihypertensive drugs might cause additive blood pressure lowering effects and increase the risk of hypotension.
Animal research shows that forskolin, a constituent of coleus, may lower blood pressure.
Cytochrome P450 2C9 (Cyp2C9) Substrates
Theoretically, taking coleus may affect drugs metabolized by CYP2C9 and increase the risk of adverse effects or reduce the effectiveness.
Research on the effect of coleus on CYP2C9 is conflicting. Some animal research shows that coleus extract can induce CYP2C9, while in vitro research shows that coleus can inhibit CYP2C9. Until more is known, advise patients that taking coleus might increase or decrease levels of drugs metabolized by CYP2C9.
Cytochrome P450 3A4 (Cyp3A4) Substrates
Theoretically, taking coleus might decrease serum levels of drugs metabolized by CYP3A4.
In vitro research shows that coleus can activate the nuclear receptor, pregnane X receptor (PXR), which results in increased expression of CYP3A4. Although the clinical significance of this is not known, use caution when considering concomitant use of coleus and other drugs affected by these enzymes.
Warfarin (Coumadin)
Theoretically, taking coleus may affect the metabolism of warfarin and increase the risk of adverse effects or reduce the effectiveness.
Some animal research shows that coleus extract can induce cytochrome P450 2C9 (CYP2C9), an enzyme that metabolizes warfarin. However, other in vitro research shows that coleus can inhibit CYP2C9. Theoretically, taking coleus with drugs metabolized by CYP2C9 might affect drug levels and the risk of adverse effects. Until more is known, advise patients that taking coleus might increase or decrease levels of warfarin.

Theoretically, chromium may have additive effects with antidiabetic agents and increase the risk of hypoglycemia.
Some research shows that taking chromium might lower blood glucose levels, especially in patients with poorly controlled type 2 diabetes.
Insulin
Theoretically, concomitant use of chromium and insulin might increase the risk of hypoglycemia.
In clinical research, chromium has been shown to increase insulin sensitivity,
Levothyroxine (Synthroid, Others)
Chromium might bind levothyroxine in the intestinal tract and decrease levothyroxine absorption.
Clinical research in healthy volunteers shows that taking chromium picolinate 1000 mcg with levothyroxine 1 mg decreases serum levels of levothyroxine by 17% when compared to taking levothyroxine alone. Advise patients to take levothyroxine at least 30 minutes before or 3-4 hours after taking chromium.
Aspirin
Theoretically, aspirin might increase chromium absorption.
Animal research suggests that aspirin may increase chromium absorption and chromium levels in the blood.
Nonsteroidal Anti-Inflammatory Drugs (Nsaids)
NSAIDs might increase chromium levels in the body.
Drugs that are prostaglandin inhibitors, such as NSAIDs, seem to increase chromium absorption and retention.
Raspberry Ketone
Stimulant Drugs
Theoretically, raspberry ketone might increase the risk of adverse cardiovascular effects with stimulant drugs.
Structurally, raspberry ketone resembles synephrine, a known stimulant agent. Heart palpitations, elevated blood pressure, coronary vasospasm, pulseless electrical activity arrest, and resistant polymorphic ventricular tachycardia have been reported in patients taking raspberry ketone.
Warfarin (Coumadin)
Theoretically, raspberry ketone might increase warfarin dose requirements.
In one case report, a patient taking warfarin 55 mg per week had a decrease in INR over a period of one month while taking raspberry ketone 250 mg daily. A warfarin dose increase to 70 mg per week was necessary to maintain a therapeutic INR while taking raspberry ketone. The mechanism for this potential interaction is not known.
