What is this page for?
First and foremost: checking L-Tryptophan 500 mg against your medications. The heart of this page is the interaction checker and the full interaction report — how this product’s ingredients may interact with prescription and over-the-counter medicines you may be taking.
Around that, we add a pharmacist’s high-level view of the product as a whole — what’s inside, the evidence for its stated use, how transparent the label is, and what safety data exists — so you can see the full picture in one place. It’s educational information from our licensed clinical databases and the clinical staff at HelloPharmacist — not medical advice — and we don’t sell or endorse products. Our editorial policy

Our pharmacy team’s full take, with four database checks built into the cards below — a summary of what is known, not a grade of the product itself.
What’s inside
Full disclosure
This supplement contains one active ingredient: L-tryptophan, an amino acid your body uses to make serotonin, a brain chemical involved in mood and sleep. The capsule also holds inactive ingredients — microcrystalline cellulose, hydroxypropyl methylcellulose, magnesium stearate, silicon dioxide, and water — which are fillers and binders that don't add therapeutic value.
Does it work?
Insufficient evidence
The evidence for L-tryptophan in depression is rated possibly ineffective based on the data we hold. For anxiety, athletic performance, teeth grinding (bruxism), and premenstrual dysphoric disorder, there isn't enough reliable evidence to rate whether it works.
If you're considering it for any of these uses, talk with your doctor about whether it's right for you.
How safe is it?
Well-documented data
L-tryptophan is generally well tolerated, though mild side effects are common: belching, diarrhea, drowsiness, dry mouth, gas, headache, heartburn, lightheadedness, nausea, stomach pain, blurred vision, and vomiting. There's an important historical note: in 1989, a contaminated batch from a single Japanese manufacturer caused over 1,500 cases of a serious neurological disorder called eosinophilia-myalgia syndrome (EMS), leading to an FDA alert and recall.
Current products are not from that source, but caution is warranted. The safety data advises against use during pregnancy and breastfeeding — there isn't enough evidence to know it's safe in either situation, so talk with your doctor or pharmacist before taking it if you're pregnant, planning pregnancy, or breastfeeding.
Meds to double-check
Major interaction found
Before taking this product, check with your doctor or pharmacist if you take any CNS depressants — sedatives, sleep aids, opioids, or other drugs that slow your nervous system — or any serotonergic drugs, including most antidepressants, some migraine medications, and certain pain relievers. These carry Major and Moderate interaction risks, respectively.
The bottom line
Scorecard at a glanceFully disclosed formula with insufficient evidence for its stated purpose. Major medication interactions have been identified, and safety information is well characterized.
L-Tryptophan may appeal to you if you're interested in a natural approach to mood or sleep, but the evidence for depression is weak and for other uses it's unproven. It's not right for you if you take CNS depressants or serotonergic drugs like most antidepressants without clearing it first.
Check with your own doctor or pharmacist before starting, especially if you're on any psychiatric or pain medication.

L-Tryptophan 500 mg by Jarrow Formulas Drug Interactions
HelloPharmacist Interaction Report
L-Tryptophan 500 mg by Jarrow Formulas interacts with two major drug categories through its amino acid content.
The most serious concern is with CNS depressants — drugs that slow your central nervous system, like sedatives, sleep aids, and opioids. Taking L-tryptophan with these can add to their drowsy effects, and clinical research shows L-tryptophan itself causes fatigue and drowsiness.
Read the full breakdown — every affected drug type, severity by severity
The product also carries Moderate-severity interactions with serotonergic drugs — medications that work on serotonin pathways, including most antidepressants, some pain relievers, and migraine medications. L-tryptophan is a building block for serotonin, so combining it with these drugs might amplify serotonin-related side effects, including a rare condition called serotonin syndrome.
Altogether, these interactions span 394 individual medications. Before you start this supplement, check your exact medications with the tool on this page or run them past your own doctor or pharmacist.
