The evidence on L-tryptophan is mixed and sparse. For depression, it's rated possibly ineffective — meaning the data doesn't support it as a treatment.

How safe is it?

Orally, L-tryptophan is generally well tolerated, though common side effects include belching, diarrhea, drowsiness, dry mouth, flatulence, headache, heartburn, lightheadedness, nausea, stomach pain, blurred vision, and vomiting. The product carries a caution because of a contamination event in the late 1980s: over 1,500 cases of a serious neurological disorder called eosinophilia-myalgia syndrome (EMS) were linked to L-tryptophan, though about 95% traced to contaminated product from a single Japanese manufacturer.
The supplement was recalled in 1990 and strict FDA limits were put in place. Modern sources are considered safer, but the history matters.
For pregnancy, there is not enough safety data — avoid it. The same applies to breastfeeding.
