Rare side effects include mild sedation, irritability, insomnia, fatigue, memory issues, headache, soft stools, nausea, vomiting, and upper GI discomfort; these resolve quickly if you stop taking it.
Lysine is generally well tolerated at typical doses, but high amounts may cause stomach upset and long-term safety isn't well studied. There isn't enough reliable information on supplement use during breastfeeding.
In pregnancy, lysine from food is fine, but supplement safety is not well studied — check with your doctor first. The most common side effects are abdominal pain, diarrhea, and indigestion.

Theoretically, glycine might decrease the effectiveness of clozapine.
One small clinical study in patients with schizophrenia shows that adding glycine to clozapine therapy worsens symptoms of schizophrenia when compared with clozapine alone. The mechanism of this interaction is unclear.

Theoretically, lysine may reduce the effects of 5-HT4 agonists.
