N.O. Support Ingredients & Drug Interactions
by GNC Mega Men
What is this page for?
First and foremost: checking N.O. Support against your medications. The heart of this page is the interaction checker and the full interaction report — how this product’s ingredients may interact with prescription and over-the-counter medicines you may be taking.
Around that, we add a pharmacist’s high-level view of the product as a whole — what’s inside, the evidence for its stated use, how transparent the label is, and what safety data exists — so you can see the full picture in one place. It’s educational information from our licensed clinical databases and the clinical staff at HelloPharmacist — not medical advice — and we don’t sell or endorse products. Our editorial policy

What’s inside
Full disclosure
GNC Mega Men N.O. Support contains one active ingredient: MegaNatural-BP Grape seed extract.
This is a concentrated extract from grape seeds, used in the supplement for its compounds called proanthocyanidins. The product also includes several inactive ingredients—cellulose, gelatin, stearic acid, magnesium stearate, and silica—which serve as the capsule material and binders.
Does it work?
Moderate evidence
The evidence for grape seed extract is mixed. It's rated possibly effective for chronic venous insufficiency (CVI)—a condition where blood pools in the veins of the legs, causing swelling and discomfort.
For other uses listed in this product, the evidence is less clear: it's rated possibly ineffective for chemotherapy-induced nausea and vomiting (CINV), allergic rhinitis (hay fever), and obesity. If you're considering this product for one of those other purposes, the science doesn't currently support its use.
How safe is it?
Well-documented data
Grape seed extract appears generally well tolerated in studies. The most common side effects are mild—abdominal pain, diarrhea, dry mouth, indigestion, headache, joint pain, and nausea.
Anaphylaxis to grape skin is rare. For pregnancy, concentrated grape supplements should be avoided due to limited safety data; whole grapes as food are fine.
For breastfeeding, there isn't enough information on concentrated grape supplements, so talk with your doctor or pharmacist if you're nursing and considering this product.
Meds to double-check
Moderate interaction found
Before taking this product, double-check with your doctor or pharmacist if you're on any blood thinners or antiplatelet drugs (like warfarin, aspirin, or clopidogrel)—grape seed extract may increase bleeding risk. Also flag it if you take cyclosporine (an immunosuppressant), midazolam (a sedative), or any medications processed by your liver's CYP2D6, CYP2E1, or CYP3A4 enzymes, which include many antidepressants, blood pressure meds, and statins.
The interactions are moderate in severity and haven't all been seen in people, but your pharmacist can confirm whether any of your specific drugs are affected.
The bottom line
Scorecard at a glanceFully disclosed formula with some supporting evidence for its stated purpose. Moderate medication interactions have been identified, and safety information is well characterized.
If you have chronic venous insufficiency and are looking for a supplement option, grape seed extract may be worth discussing with your doctor. But if you take blood thinners, antiplatelet drugs, cyclosporine, midazolam, or certain liver-processed medications, you'll need to check with your doctor or pharmacist before adding this product—the interactions are real and could affect how your drugs work or increase bleeding risk.
Start by running your medications through the checker on this page.

N.O. Support by GNC Mega Men Drug Interactions
HelloPharmacist Interaction Report
GNC Mega Men N.O.
Support contains grape seed extract, which has documented interactions with a number of medications. The most serious concern is with blood thinners and antiplatelet drugs (medications that prevent blood clots)—grape seed extract may increase bleeding risk if you're taking these.
It theoretically has antiplatelet effects and in lab studies shows decreased platelet aggregation, though human cases haven't been reported yet.
Read the full breakdown — every affected drug type, severity by severity
Grape seed extract may also affect how your body processes several classes of drugs. It can theoretically increase levels of certain medications that are broken down by liver enzymes called CYP2D6 and CYP2E1 (including some antidepressants and pain relievers), though again, this hasn't been seen in people.
Its effect on CYP3A4 substrates—a large group that includes some statins, blood pressure meds, and immunosuppressants—is unclear; research suggests it could either increase or decrease their levels.
Two specific drugs warrant extra caution. If you take cyclosporine (an immunosuppressant), grape seed extract can reduce its absorption by up to 30%; separate your doses by at least 2 hours.
If you take midazolam (a sedative), long-term grape seed extract use might decrease how well it works, though a single dose appears safe. Phenacetin (an older pain reliever, rarely used today) levels may also drop.
Additionally, long-term use could theoretically reduce levels of certain CYP1A2 substrates, though one small study of grape juice showed it actually decreased phenacetin levels. Altogether, these interactions span 911 individual medications.

MegaNatural-BP Grape seed extract
Anticoagulant/Antiplatelet Drugs
Theoretically, grape extracts may have antiplatelet effects and may increase the risk of bleeding if used with anticoagulant or antiplatelet drugs.
In vitro evidence suggests that grape extracts might decrease platelet aggregation.
Cyclosporine (Neoral, Sandimmune)
Ingesting grape juice with cyclosporine can reduce cyclosporine absorption.
A small pharmacokinetic study in healthy young adults shows that intake of purple grape juice 200 mL along with cyclosporine can decrease the absorption of cyclosporine by up to 30% when compared with water. Separate doses of grape juice and cyclosporine by at least 2 hours to avoid this interaction.
Cytochrome P450 1A2 (Cyp1A2) Substrates
Theoretically, grape juice might reduce the levels of CYP1A2 substrates.
A small pharmacokinetic study in healthy adults shows that ingestion of 200 mL of grape juice decreases phenacetin plasma levels. This is thought to be due to induction of CYP1A2.
Cytochrome P450 2D6 (Cyp2D6) Substrates
Theoretically, grape seed extract may increase the levels of CYP2D6 substrates.
In vitro evidence suggests that grape seed extract might inhibit CYP2D6 enzymes. However, this interaction has not been reported in humans.
Cytochrome P450 2E1 (Cyp2E1) Substrates
Theoretically, grape seed extract might increase the levels of CYP2E1 substrates.
In vitro and animal research suggests that grape seed proanthocyanidin extract inhibits CYP2E1 enzymes. However, this interaction has not been reported in humans.
Cytochrome P450 3A4 (Cyp3A4) Substrates
It is unclear if grape seed extract inhibits or induces CYP3A4; research is conflicting.
In vitro evidence suggests that grape seed extract might inhibit CYP3A4 enzymes. However, evidence from animal research shows that grape seed extract may induce CYP3A4 in the liver. So far, these interactions have not been reported in humans.
Midazolam (Versed)
Theoretically, long-term intake of grape seed extract might decrease the effects of midazolam.
Animal research shows that subchronic ingestions of grape seed extract can increase the elimination of intravenous midazolam by increasing hepatic CYP3A4 activity. Single doses of grape seed extract do not appear to affect midazolam elimination.
Phenacetin
Grape juice might decrease phenacetin absorption.
A small pharmacokinetic study in healthy adults shows that ingestion of 200 mL of grape juice decreases phenacetin plasma levels. This is thought to be due to induction of cytochrome P450 1A2 (CYP1A2).
Cytochrome P450 2C9 (Cyp2C9) Substrates
It is unclear if grape juice or grape seed extract inhibits CYP2C9; research is conflicting.
In vitro evidence shows that grape seed extract or grape juice might inhibit CYP2C9 enzymes. However, a small pharmacokinetic study in healthy adults shows that drinking 8 ounces of grape juice once does not affect the clearance of flurbiprofen, a probe-drug for CYP2C9 metabolism. The effects of continued grape juice consumption are unclear.
