Papain is generally tolerated as a food, but supplemental forms may irritate your digestive tract and trigger allergic reactions in sensitive people. Short-term use at normal doses is well tolerated orally; high doses may be unsafe and can rarely cause severe stomach inflammation or esophageal perforation.

Occupational exposure to papain dust can cause breathing problems, sneezing, eye irritation, and asthma symptoms in those sensitive to it. During pregnancy, concentrated papain and unripe papaya products are best avoided due to safety concerns.
