The evidence for L-tryptophan is mixed. For depression, the data suggests it's possibly ineffective.
For anxiety, athletic performance, teeth grinding (bruxism), and premenstrual dysphoric disorder, there isn't enough reliable evidence in our data to say whether it works.
How safe is it?
Well-documented data
L-Tryptophan is generally well tolerated when taken by mouth, but it does come with a cautionary history. Common side effects include belching, diarrhea, drowsiness, dry mouth, flatulence, headache, heartburn, lightheadedness, nausea, stomach pain, blurred vision, and vomiting.
In the late 1980s, a contaminated batch of L-tryptophan from a single Japanese manufacturer caused over 1,500 cases of a serious neurological disorder called eosinophilia-myalgia syndrome (EMS); the FDA issued a recall and import limits in 1990. Modern products are not known to carry this risk, but the history warrants awareness.
There isn't enough safety data to use L-tryptophan during pregnancy — talk with your doctor or pharmacist about whether it's right for you. It should be avoided while breastfeeding because safety hasn't been established.
