Most of these amino acids are generally well tolerated at typical doses, though long-term safety data are limited for several of them. The most commonly reported side effects across the ingredients are gastrointestinal: nausea, vomiting, diarrhea, abdominal pain, bloating, and dyspepsia.

Glycine has rarely caused mild sedation, and phenylalanine may cause anxiety, insomnia, headache, heartburn, or nausea. Methionine can cause dizziness, drowsiness, and increased homocysteine (a risk factor for blood vessel problems), especially at high doses.

Serine at very high doses has caused seizures. Regarding pregnancy and breastfeeding: aspartic acid and phenylalanine are not recommended in supplement doses during pregnancy or breastfeeding; serine, tyrosine, and glutamine should be avoided in supplement amounts unless a doctor advises otherwise; glycine, methionine, lysine, threonine, histidine, and proline lack sufficient safety data, so discuss with your healthcare provider before use.

Phenylalanine competes with levodopa for carrier-mediated transport into the brain. The resulting reduction in levels of levodopa in the brain can exacerbate tremor, rigidity, and the "on-off" phenomenon in patients with Parkinson disease.

Concomitant intake of phenylalanine may reduce the intestinal absorption of baclofen.
Phenylalanine and baclofen share the same intestinal carrier for absorption; phenylalanine competitively inhibits the absorption of baclofen, reducing its plasma levels.

Monoamine Oxidase Inhibitors (Maois)

Theoretically, concomitant use of L-phenylalanine and non-selective MAOIs might increase the risk of hypertensive crisis.
L-phenylalanine is metabolized to tyrosine. Some evidence suggests that L-phenylalanine, given with the non-selective MAOI pargyline, might prevent the elimination of tyramine, increasing the risk of hypertensive crisis. However, this was not reported in a small number of patients when using L-phenylalanine with the partially selective MAO-B inhibitor, selegiline.

Phenylalanine in this blend competes with levodopa for transport into your brain, which can reduce the drug's effectiveness and worsen tremor and stiffness. The two should be separated by at least 2 hours, but that's a conversation for your doctor or pharmacist.
