Amiodarone is a CYP2C8 substrate1. Abiraterone acetate inhibits CYP2C8 and increased the AUC of pioglitazone (a CYP2C8 probe substrate) by 46% when coadministered with a single dose of abiraterone acetate 1000 mg 3. Avoid coadministration of amiodarone with a CYP2C8 inhibitor as exposure to amiodarone may be increased. Due to the long half-life of amiodarone, drug interactions may persist for weeks to months after discontinuation 1. Monitor patients for signs of toxicity related to a CYP2C8 substrate such as amiodarone for which a minimal change in plasma concentration may lead to serious or life-threatening adverse reactions 2.
