Asciminib is a CYP3A4 substrate. Concomitant use of asciminib with a strong CYP3A4 inhibitor may increase asciminib exposure, which may increase the risk of asciminib-related adverse reactions. The asciminib AUC (inf) and Cmax increased by 36% and 19%, respectively, following coadministration of a single asciminib dose of 40 mg with a strong CYP3A4 inhibitor (clarithromycin). Closely monitor patients for adverse reactions in patients treated with asciminib at 200 mg twice daily with concomitant use of strong CYP3A4 inhibitors1.
