Asciminib falls under CYTOCHROME P450 3A4 (CYP3A4) INHIBITORS — the group Ergot interacts with.

Ergotamine, a constituent of ergot, is a substrate of cytochrome P450 3A4 (CYP3A4). Theoretically, drugs that inhibit CYP3A4 might increase the risk of ergot toxicity. Some of these drugs include amiodarone (Cordarone), clarithromycin (Biaxin), diltiazem (Cardizem), erythromycin (E-mycin, Erythrocin), indinavir (Crixivan), ritonavir (Norvir), saquinavir (Fortovase, Invirase), and many others.

Here's the key thing to know: your leukemia medicine asciminib (Scemblix) can mildly block the liver enzyme CYP3A4, which is the same enzyme that clears ergotamine out of your body. When that enzyme is slowed, ergot can build up and become more concentrated. Ergot is already a strong vessel-narrowing chemical, so higher levels can mean dangerous effects like cold or numb hands and feet, leg cramps, severe headache, or chest pain.

I'll be straight with you though: this is based on weak, anecdotal evidence and is partly theoretical. The much bigger issue is that crude ergot is toxic and unpredictable on its own and really shouldn't be used as a home supplement at all. Please run anything ergot-related past your oncologist or pharmacist first.

Ergot is a toxic fungus that grows on rye and contains powerful chemicals (ergot alkaloids) that strongly narrow blood vessels and squeeze the uterus. Because raw ergot is poisonous and the dose is impossible to control safely, it should never be used as a home remedy or supplement. Some purified ergot chemicals are used as prescription drugs under strict medical supervision, but the crude herb is not safe to take.

What Is Asciminib Used For?

Brand name: Scemblix

Scemblix (asciminib) is used to treat adult patients with Philadelphia chromosome-positive chronic myeloid leukemia in chronic phase (Ph+ CML-CP) — a type of blood cancer driven by an abnormal protein called BCR-ABL1. It covers three groups: people newly diagnosed with Ph+ CML-CP, people who have already been treated for Ph+ CML-CP, and people whose cancer carries a specific mutation called T315I that makes it harder to treat.

The use in newly diagnosed Ph+ CML-CP was approved under accelerated approval based on early response data. Continued approval for that specific use may depend on results from confirmatory trials confirming long-term clinical benefit.
