Increased atorvastatin exposure

Interaction Deep Dive

Coadministration of atorvastatin, a CYP3A4 substrate and transporters of P-gp1 and boceprevir (inhibitor of CYP3A4 and P-gp) increased atorvastatin exposure. If these agents are coadministered, use the lowest effective dose of atorvastatin. However, do not exceed atorvastatin 40 mg daily when given concurrently with boceprevir 2.

a) In a drug interaction study in healthy volunteers or patients with hepatitis C virus (HCV) positive genotype-1, coadministration of a single dose of atorvastatin 40 mg and boceprevir 800 mg three times daily for 7 days resulted in increased Cmax and AUC. The ratio estimate (in combination with boceprevir to atorvastatin alone) was 2.66 (90% CI, 1.81 to 3.9) for Cmax and 2.30 (90% CI, 1.84 to 2.88) 21.
