Concomitant use of asciminib (a CYP3A4 inhibitor) with a CYP3A4 substrate increases the exposure of the CYP3A4 substrate, which may increase the risk of adverse reactions. Coadministration of asciminib with midazolam (a CYP3A4 substrate) has resulted in an increase in midazolam exposure with increases of 24% to 88% in AUC(inf) and by 11% to 58% in Cmax. Avoid coadministration of asciminib 200 mg twice daily with a CYP3A4 substrate where minimal concentration changes may lead to serious adverse reactions. In patients receiving a total dose of asciminib 80 mg/day, closely monitor for adverse reactions when coadministered with such a CYP3A4 substrate where minimal concentration changes may lead to serious adverse reactions1.
