Concomitant administration of piperaquine and QT-interval prolonging drugs may result in additive prolongation effects on the QT interval and is contraindicated. Additionally, recent use of QT-interval prolonging drugs, that may still be circulating at the time of piperaquine administration, is contraindicated. Concurrent administration of piperaquine (a CYP3A4 inhibitor) and a CYP3A4 substrate may increase the exposure of the CYP3A4 substrate. Due to the long half-life of piperaquine, caution is advised when administering a CYP3A4 substrate for up to 3 months after discontinuation of piperaquine therapy1.
