Avoid coadministration of sotorasib with proton pump inhibitors and H2-antagonists because reduced sotorasib exposure may result in diminished anti-tumor activity. In a pharmacokinetic study, coadministration of sotorasib with omeprazole (proton pump inhibitor) significantly reduced sotorasib exposure when given with and without food. In another study, coadministration with famotidine (H2-antagonist), separated by 10 hours prior or 2 hours after sotorasib also reduced sotorasib exposure1.
