St. John's wort induces CYP3A4 enzymes and increases metabolism of CYP3A4 substrates. Clinically significant interactions have been reported with St. John's wort products containing hyperforin 1 mg or more.

Here's the concern with pairing these two. St. John's wort revs up a liver enzyme called CYP3A4, and your body uses that same enzyme to break down Sotorasib (Lumakras). When the enzyme works faster, your sotorasib gets cleared out of your system more quickly, which can drop its blood levels.

This matters a lot because sotorasib is a cancer treatment, and lower levels can mean it doesn't fight your lung cancer as well as it should. This isn't about more side effects, it's about losing the benefit you need. The evidence here is solid enough that I'd treat it as a real problem. Please don't stop either one on your own, but do talk with your oncologist or pharmacist before combining them so they can keep your treatment working.

What Is Sotorasib Used For?

Brand name: Lumakras

Lumakras is used to treat adults with KRAS G12C-mutated locally advanced or metastatic non-small cell lung cancer (NSCLC) who have already received at least one prior cancer treatment. This use was approved based on tumor response data, and continued approval depends on results from ongoing confirmatory studies.

Lumakras is also approved — used together with panitumumab — for adults with KRAS G12C-mutated metastatic colorectal cancer (mCRC) who have already received prior chemotherapy regimens containing fluoropyrimidine, oxaliplatin, and irinotecan. In both cases, patients must have a confirmed KRAS G12C mutation detected by an FDA-approved test before starting treatment.
