Coadministration of pirtobrutinib and digoxin (a P-gp substrate with NTI) increased the exposure of digoxin, which may increase the risk of adverse reactions related to P-gp substrates for drugs which are sensitive to minimal concentration changes. In a drug interaction study, a single 200 mg dose of pirtobrutinib increased the AUC and Cmax of digoxin (a sensitive P-gp substrate) by 17% and 51%, respectively. Multiple doses of pirtobrutinib (200 mg daily) further increased the AUC and Cmax of digoxin up to 35% and 55%, respectively3.
