Reduced dasatinib absorption, reduced efficacy of dasatinib, increased dasatinib exposure and an increased risk of dasatinib-related adverse and toxicity

Interaction Deep Dive

Vonoprazan reduces intragastric acidity, which may reduce the absorption of dasatinib reducing the effectiveness of dasatinib²³. Also, concomitant administration of vonoprazan (a CYP3A4 inhibitor) and dasatinib (CYP3A4 substrate with NTI) may result in increased exposure of dasatinib where minimal concentration changes may lead to serious toxicities. Frequently monitor concentrations and/or adverse reactions related to dasatinib. Dosage reduction of dasatinib may be needed ⁴. Do not administer proton pump inhibitor ie., vonoprazan with dasatinib. Alternatively, use antacid at least 2 hours prior to or 2 hours after the dose of dasatinib ¹.
