Polatuzumab Vedotin-Piiq falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Coleus interacts with.
In vitro research shows that coleus can activate the nuclear receptor, pregnane X receptor (PXR), which results in increased expression of CYP3A4. Although the clinical significance of this is not known, use caution when considering concomitant use of coleus and other drugs affected by these enzymes.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the gist with Polivy (polatuzumab vedotin-piiq) and Coleus. This medicine is an antibody-drug conjugate that delivers a potent cancer-killing payload (called MMAE) into lymphoma cells, and that payload is partly broken down by a liver enzyme named CYP3A4. Lab studies hint that Coleus may rev up CYP3A4, which could theoretically clear the drug a little faster and lessen how well it works.
I want to be honest: this is theoretical, based only on test-tube data, so we don't really know if it matters in real people. Still, since Polivy is serious chemo for a hard-to-treat lymphoma, I wouldn't gamble with it. Please don't start Coleus without first running it by your oncologist or pharmacist.

Polatuzumab vedotin-piiq injection is used along with bendamustine (Belrapzo, Treanda) and rituximab (Rituxan) in adults to treat a certain type of non-Hodgkin's lymphoma (NHL; a type of cancer that begins in a type of white blood cells that normally fights infection) that did not improve or improved but returned after treatment with at least two other chemotherapy medications. Polatuzumab vedotin-piiq is in a class of medications called antibody-drug conjugates. It works by killing cancer cells.
