Delta-9-tetrahydrocannabinol (thc) and Trofinetide

Here is the interaction we found between these two, with the evidence behind it.

Moderate

CYTOCHROME P450 3A4 (CYP3A4) INHIBITORS

Why this applies to Trofinetide

Trofinetide falls under CYTOCHROME P450 3A4 (CYP3A4) INHIBITORS — the group Delta-9-tetrahydrocannabinol (thc) interacts with.

Theoretically, CYP3A4 inhibitors might increase the levels and clinical effects of THC.
THC is a substrate of CYP3A4 enzymes.
Severity Moderate Likelihood Possible Evidence D

Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.

Dr. Brian Staiger, PharmD, BCPS
Dr. Brian Staiger explains PharmD, Clinical Pharmacist

Here's the gist for this pairing. Your body uses an enzyme called CYP3A4 to help clear THC. Trofinetide (Daybue) can act as a mild blocker of that enzyme, so in theory it could slow down how fast THC leaves your system. When THC lingers, its effects can feel stronger or last longer, meaning more of that familiar high, along with drowsiness, dizziness, a racing heart, or feeling foggy or off balance.

I want to be honest that this concern is theoretical, based on how these two behave in the lab rather than real patient reports, so it may never cause a noticeable problem. Still, since THC affects thinking and driving, it's worth a quick chat with your pharmacist or doctor before combining them, especially for a child on Daybue.
